Týdenní praktický kurz je určen k seznámení studentů s metodami virové biotechnologie. Studenti získají
teoretické základy o základních principech virových expresních systémů, jejich možnosti využití pro expresy
rekombinantních proteinů nebo pro produkci virových částic a metodách jejich izolace. S využitím virových částic
jako modelu získají studenti praktické zkušenosti s metodami produkce (práce s buněčnými kulturami a
rekombinantními viry) a izolace (centrifugace v gradientech hustoty) virových částic.

One-week practical course is designed to introduce students to the methods of viral biotechnology. Students gain
strong theoretical background about the basic principles of viral expression systems, their possibility of usage for
recombinant proteins expression or for production of virus-like particles and methods of their purification. Using
virus-like particles as a model, the students gain practical experience with the methods of production (work with
cell cultures and recombinant viruses) and purification (centrifugation in density gradients) of virus like particles.

For passing the course, the following requirements must be fulfilled:
Participate actively in the course; any unexcused absence may be reason for disqualification from the course.
During the course, focus only on activities related to the course.
Submit protocols and answers to control questions due to date specified. This text can be returned for revision only once after consultation, credit will not be granted in case of repeated deficiencies.

Students will be diveded into groups (2 students per group) and each group will isolate VLPs by three types of centrifugation - differencial and zonal centrifugation, isopycnic centrifugation. Purity of the VLPs will be characterized using electron microscopy technique.

After completing the course, the student will:
•Describe the basic principle of viral expression system (with emphasis on the baculovirus expression system) and discuss their usage in producing of VLPs.
•Explain the principle of different centrifugation methods and compare their advantages and disadvantages in context of VLPs isolation.
•Independently perform VLPs isolation, purification and characterization by electron microscopy.
•Be able to analyse and interpret experimental results, including the evaluation of electrophoretic records, and clearly process them in the form of a laboratory protocol.
•Discuss the most reliable centrifugation method for VLPs isolation according to data obtained during the course.
•Design own protocol for isolating VLPs.

Vstupní požadavky - angličtina
· The course starts on Monday, 23.3.2026 (preliminary date) at 9.00 am in BIOCEV building (Prumyslova 595; Vestec), laboratory L1-012. Meeting point is at reception at 9:00. Enrolled participants are allowed to enter the laboratory only in laboratory clothing and shoes.
