Přednáška je zaměřena na osvojení si základních znalostí o biochemických procesech vedoucích k vývoji nádorových onemocnění. Tento kurz je určen pro studenty chemických oborů, především biochemikům. Příčiny a průběh nádorových onemocnění jsou studentům vysvětleny z hlediska molekulární podstaty těchto chorob. Detailně jsou studenti informováni o faktorech vyvolávajících a ovlivňujících procesy karcinogenese a o mechanismu základních fází procesu karcinogenese (iniciace, promoce, progrese). Z biochemického hlediska jsou vysvětleny testy sledující karcinogenitu chemických sloučenin (dlouhodobé a krátkodobé testy na karcinogenitu). Při osvětlení procesů iniciační fáze karcinogenese jsou studenti seznamováni s reakcemi vedoucími k aktivaci chemických karcinogenů a enzymy katalysujícími takové reakce, dále pak s mechasnismy tvorby prekarcinogenních lézí DNA (aduktů v DNA). V promoční fázi karcinogene je podtržena funkce aktivních forem kyslíku, radikálů a proteinkinasy C na potenciaci dělení buněk. Progresí fáze karcinogenese je vysvětlena jako finální genotoxický zásah do buněk prekarcinogenních (benigních) lézí. Dosud vědecky nevysvětlená orgánová specifika karcinogenních faktorů je uvedena jako příklad oblasti hodné detailního vědeckého bádání. Velký důraz je v kursu kladen na principy léčby nádorových onemocnění (chemoterapie, radiační terapie, chirurgické zákroky). Protinádorová chemoterapeutika jsou klasifikována podle mechanismu jejich působení (antimetabolity, interkalátory, alkylační/arylační činidla, mitotické jedy, deriváty hormonů, inhibitory synthesy hormonů, inhibitory topoisomeras, inhibitory proteosynthesy). Podtržen je i význam epidemiologických studií a prevence ve snížení nádorových onemocnění.

The aim of the course is to explain the biochemical processes leading to development of cancer diseases, their treatment and prevention. The course is a lecture useful for chemists and especially for biochemists. At first, essential information on cancer as a disease is given (tumors, their classification and characterization), on cancer development, factors inducing and influencing the processes of carcinogenesis. The mechanism of basic phases of carcinogenic processes / initiation, promotion, progression is explained. Data on metastasis processes are also given. Three phases of carcinogenesis are shown in details (initiation developed from factors causing the DNA damage, promotional phase - increased proliferation, mechanism of the effects of radicals and the active forms of oxygen, enzymes participating in imitational and promotional phases of carcinogenesis, the function of protein kinase C). Progression phase of carcinogenesis is shown as the phase involving additional, final, modification of DNA. The role of endogenous and exogenous carcinogens and enzymes participating in this phase of carcinogenesis is underlined. The target of the course is also to explain the organ specificity of carcinogens and repair systems for the DNA lesions. Final parts of the course are aimed at explanation of principles of cancer treatment (chemotherapy, radiation, surgery), in details are classified anticancer drugs and their mechanism of action (antimetabolites, intercalators, drug alkylating/arylating DNA, mitotic drugs, hormone derivatives, inhibitors of hormone synthesis, inhibitors of topoisomerases, inhibitors of protein synthesis). The significance of epidemiology cancer prevention is underlined.
