Cílem je podat úvod do toxikokinetiky a farmakokinetiky, včetně zopakování a prohloubení základních potřebných znalostí z obecné toxikologie a z fyzikální chemie. V přednášce jsou probrány základní toxikokinetické procesy, související s dostupností účinné chemikálie organizmu. Na nich jsou vyloženy základní pojmy toxikokinetiky, používané modely jedno- a dvoukompartmentové a kinetické simulační fyziologické modely. Je probrána závislost velikosti biologického účinku na dávce (koncentraci), na čase působení a na chemické struktuře chemikálií. Extrapolace mezi daty, poskytovanými různými biologickými druhy nebo získanými po různém typu podání jsou diskutovány při využití toxikokinetiky. Podrobněji je vyložen kvantitativní odhad rizik expozice chemikáliím a jeho využití pro zdraví přírody i člověka. Využití toxikokinetických znalostí je ukázáno při výpočtu účinné dávky, na tvorbě biologických expozičních testů a interpretaci jejich výsledků, při studiu toxických účinků směsí chemikálií. Na seminářích jsou probírány potřebné výpočty modelů a jejich použití.

The aim is to give an introduction to toxicokinetics and to pharmacokinetics involving repetition of knowledge from general toxicology and physical chemistry necessary for toxikokinetics. The course covers the basic toxicokinetic processes in relation to a bioavailability of a chemical to organisms. The usage of one- and two-compartment models and kinetic simulation physiology-based models in toxicokinetic studies is shown. The dependence of magnitude of biological effect on dose (concentration), on exposure duration and on chemical structure of effective chemicals is discussed. Based on the knowledge gained, extrapolation among data obtained with various tested biological objects or after various routes of application a chemical are interpreted in terms of toxicokinetics. Quantitative assessment of risk of chemical exposure and its usage for a health of nature and a human is discussed in details. The usage of toxicokinetic knowledge is shown in a calculation of an effective dose, in a development and use of biological exposure tests and interpretation of their results or in studies of toxic effects of chemical mixtures.The seminar is devoted to a calculation and usage of the models.

Požadavky ke zkoušce

Zápočet se uděluje na základě úspěšně napsaného testu zaměřeného na výpočty (minimální hranice je 65% bodů z možných). Zkouška probíhá, po získání zápočtu, ústní formou. Zkouší se vše, co bylo odpřednášeno.

1. Terminology, terms and definitions. Knowledge of basic and general toxicilogy and chemical laws is necessary.
2. Bioavailability - toxicokinetic processes: absorption, transport, distribution, biotransformation, elimination; site of action.
3. Relationships between magnitude of a biological effect and concentration/dose (dose /concentration - response curves) and time. Relationships between chemical structure and biological activity.
4. Compartmental modelling and its use for quantitative evaluation of an exposure (clearance, area under curve, etc.)One- and two-compartmental models, calculation of kinetic parameters. Kinetics of the first and the zeroth order. Simulation of various routes of application.
5. Physiologically-based kinetic models. Simulation of physiological parameters of organisms and of physicochemical properties of an active chemical.
6. Mutual extrapolation of data obtained with various biological objects, extrapolation of data obtained with various routes of exposure, interupted exposure included.
7. Hazard and risk. Quantitative assessment of risk of exposure to chemicals: hazard identification, exposure measurement, dose(concentration )-response curve establishment, characterization of population exposed.Influence of the exposure to chemicals on environmental and human health.
