Adaptace na chlad je doprovázena změnami v tukové tkáni, a to včetně změn v imunitním systému přítomném v tukové tkáni. Počátek chladové expozice je spojen se stresovou, spíše zánětlivou reakcí, pozdější změny jsou spojovány spíše s protizánětlivým nastavením. Je tedy možné očekávat změny ve fenotypu a funkci mezenchymálních kmenových buněk (MSC) derivovaných z tukové tkáně. Cílem diplomové práce je studium těchto změn, se zaměřením na modifikace v jejich metabolismu. Dalším cílem práce je studium imunomodulačních a regeneračních vlastností MSC izolovaných z myší vystavených chladu a jejich srovnání s kontrolní skupinou. Pochopení změn ve fenotypu chladových MSC umožní jeho navození v in vitro podmínkách.

Adaptation to cold is accompanied by changes in adipose tissue, including changes in the immune system present in the adipose tissue. Early cold exposure is associated with a stress, rather inflammatory response, while later changes are associated with an anti-inflammatory setting. Thus, changes in the phenotype and function of adipose tissue-derived mesenchymal stem cells (MSCs) might be expected. The aim of this thesis is to study these changes, focusing on specific modifications in their metabolism. Another aim of the thesis is to study the immunomodulatory and regenerative properties of MSCs isolated from mice exposed to cold and to compare them with a control group. Understanding the changes in the phenotype of cold-exposed MSCs will allow its induction under in vitro conditions.
