Při použití řízených genových manipulací u modelových mechorostů Physcomitrium patens a Marchantia polymorpha tato disertační práce směřuje nejen k porozumění funkční integrace komplexů exocyst - řídí závěrečné fáze sekrece - a TTP - podílí se na regulaci dynmiky zvl. mikrotubulárního cytoskeletu jak při buněčném dělení, tak v interfázi na buněčné morfogenezi - ale také k pochopení jejich evoluce. U vybraných mutantů mechorostů bude testována funkčnost homologů příbuzných streptofytních řas. |

Land plant morphogenetic machinery evolved stepwise from the streptohype/charophytes algal origins and model mosses Physcomitrium and Marchantia are currently well established models to study cell proliferation and morphogenesis in this sister clade to tracheophytes. Using targeted gene manipulation we aim to address how exocyst complex (regulates final step of secretory pathway) and TTP complex (regulates dynamics of microtubular cytoskeleton
in cell division and interphase morphogenesis) function in proliferative and morphogenetic cell processes in model mosses and if moss genes might be complemented by homologs from streptophyte algal clades. Implementation of this dissertation should contribute not only to the understanding of functional integration of exocyst and TTP complexes into the moss cell machineries, but also to their evolution. |
