The nested case-control design (Thomas 1977) is a sampling technique used in epidemiological studies with mortality or disease-incidence outcomes. In this design, data on the follow-up and covariates of the cases is are completely known. At each observed failure time, a small number of controls is sampled from all subjects who have been at risk at that moment, and the covariates are ascertained for them. Thus, it is an example of incomplete covariate data regression problem, with missing-by-design features.
It turns out that the nested case-control design can be analyzed by unweighted partial likelihood (Goldstein and Langholz 1992), similarly to the classical Cox model. However, there are many interesting issues regarding variance estimation and efficiency improvements. The thesis will describe selected methods for the analysis of the nested case-control design and study their theoretical properties. Selected procedures extending the basic partial likelihood estimator may be discussed. The theoretical developments will be accompanied by results of simulation studies comparing the performance of various approaches on real data. |
