Model tryptofanové deplece využívá možnosti ovlivnit biologickou dostupnost esenciální aminokyseliny tryptofanu v potravě. Tryptofan je dietním prekurzorem důležitého neurotransmiteru serotoninu (5-hydroxytryptaminu). 5-hydroxytryptamin hraje významnou roli v mnoha fyziologických a behaviorálních funkcích organismu a tím také v patogenezi a patofyziologii zejména depresivních, a afektivních poruch a onemocnění. Velký význam v této oblasti je přikládán také glutamátu, který ovlivňuje synaptickou plasticitu, paměť a učení. Podáním antagonistů lze ovlibnit funkci glutamátových drah a tím i funkci drah dopaminergních. Kombinace tryptofanové deplece s antagonisty N-metyl D-aspartátových receptorů, v animálním modelu, věrněji simulují příznaky psychiatrických poruch a onemocnění. Čím dojde ke zvýšení validity modelu.

The combination of tryptophan depletion with NMDA antagonists adminsitration more better simulates the symptoms of psychiatric diseases. This increases the validity of the model.
