Somatic hybridization is a method for preparing plant hybrids based on the fusion of isolated protoplasts. This method was introduced in the 70s of the 20th century and has been used mainly for the hybridization of plants that can not interbreed in other ways. Somatic hybridization was replaced by methods of targeted gene transfer in the 80s and 90s (GMO biotechnology). This thesis is focused on the review of methodological approaches of somatic hybridization in plants. Individual chapters discuss fundamental steps such as material preparation, protoplast isolation, protoplast fusion, cell regeneration, regeneration of plants and selection of hybrid offspring.
