Fosforylace je jednou z nejrozšířenějších a nejpodstatnějších post-translačních modifikací proteinů. Konformační změna proteinu po navázání fosfátu do fosforylačního místa je samotným principem regulace. Jedním z přístupů k analýze fosfoproteinů je detekce fosforylace pomocí srovnání jejich hmotnostních spekter před a po defosforylaci fosfatázou. Opakovatelně použitelné fosfatázy imobilizované na magnetické částice zvyšují ekonomičnost analýzy, navíc nedochází ke kontaminaci vzorku samotnou fosfatázou. Cílem této bakalářské práce je nalézt podmínky vhodné pro defosforylaci bílkovin imobilizovaným enzymem. Předběžná náplň práce v anglickém jazyce
Protein phosphorylation is involved in many important cellular events; investigation and characterization of proteins involved in these processes as well as understanding a role of the protein modification represent still a difficult task. Several experimental strategies can be employed in the phosphoproteomic studies. An application of the phosphatase treatment and a comparison of the behavior of phosphorylated and dephosphorylated protein represents on of such approaches. The use of enzyme immobilized to magnetic carriers has several advantages as compared with an application of soluble forms of enzymes: an increased stability of enzymes, an easy removal of modifying enzyme from the reaction mixture, a possibility of direct use of enzyme reaction products for MS analysis, or for a further reaction, e.g. proteolytic digestion. Subject of Thesis concerns the characterization of phosphatase immobilized to magnetic beads. The ?properties of immobilized phosphatase will be compared with those of the soluble enzyme.
