Mezi civilizační choroby patří například obezita, diabetes mellitus, revmatoidní artritida, onkologické a kardiovaskulární onemocnění. Jejich výskyt v populaci se každoročně zvyšuje. Některá z těchto onemocnění jsou spojená s autoimunitními a zánětlivými stavy, které vedou k uvolňování DNA do oběhu. Zvýšené hladiny cirkulující volné DNA (cfDNA) a změny v zastoupení jejích sekvencí odrážejí progresi onemocnění a jejich analýza může sloužit ke stanovení prognózy a monitorování odpovědi na léčbu. Cílem práce je shrnout současné poznatky o cfDNA v souvislosti s patogenezí nejčastějších civilizačních nemocí se zaměřením na diabetes mellitus a obezitu, a prozkoumání možnosti jejího využití v rámci diagnostiky, monitorování průběhu nemoci a prevence.

Civilization diseases include obesity, diabetes mellitus, rheumatoid arthritis, cancer and cardiovascular diseases. Their prevalence in the population is increasing every year. Some of these diseases are associated with autoimmune and inflammatory conditions that lead to the release of DNA into the circulation. Elevated levels of circulating free DNA (cfDNA) and changes in the abundance of its sequences reflect disease progression, and their analysis can be used to determine prognosis and monitor response to treatment. The aim of this study is to summarize the current knowledge of cfDNA in the context of the pathogenesis of the most common diseases of civilization, with a focus on diabetes mellitus and obesity, and to explore its potential use in diagnosis, monitoring of disease progression and prevention.
