Cílem diplomové práce bude objasnit vztah mezi cirkadiánním systémem a modifikací RNA, jakou je metylace adenosinu (m6A). Tato modifikace ovlivňuje translaci, přesun RNA z jádra či degradaci RNA. Práce se zaměří na fungování cirkadiánního systému v savčím mozku po umlčení jednotlivých enzymů zprostředkujících m6A metylaci (metyltransferáz, demetyláz, apod.). Pro experimenty budou použity primární kultury izolované z mozkové tkáně geneticky modifikovaných myší s Per2::Luc genem, který umožňuje sledovat expresi hodinového genu Period2 v aktuálním čase.

The aim of the diploma thesis will be to clarify the relationship between the circadian system and RNA modification, such as methylation of adenosine (m6A). This modification affects the translation, transfer of RNA from the nucleus, or RNA degradation. The work will focus on the function of the circadian system in the mammalian brain after the silencing of selected enzymes mediating m6A methylation (methyltransferases, demethylases, etc.). Primary cultures isolated from the brain tissue of genetically modified mice with the Per2:: Luc gene, which allows monitoring the expression of the Period2 clock gene at the current time, will be used for the experiments.
