Diagnostika Alzheimerovy nemoci je v současné době založena na stanovení přítomnosti specifických proteinů v mozkomíšním moku, případně jejich zobrazení pomocí pozitronové emisní tomografie. Obě tyto metody jsou invazivní a mohou pacienta vystavit jistému riziku a dyskomfortu. Krevní biomarkery proto představují naději pro včasnou diagnostiku a monitoraci jedinců ve zvýšeném riziku rozvoje Alzheimerovy nemoci. Cílem diplomové práce je stanovit biomakery v krvi pomocí ultrasenzitivní Simoa TM (Single molecular assay) technologie a ověřit jejich vztah k hodnotám v mozkomíšním moku. Dále je cílem vysledovat vztah mezi krevními biomarkery a kognitivními a strukturálními markery Alzheimerovy nemoci. Výzkumný soubor budou tvořit pacienti Kognitivního centra Neurologické kliniky 2.lékařské fakulty Univerzity Karlovy, definovaní dle aktuálních AT(N) diagnostických kritérií (Jack et al., 2018).

The diagnosis of Alzheimer's disease is currently based on the determination of the presence of specific proteins in cerebrospinal fluid or their imaging using positron emission tomography. Both of these methods are invasive and can expose the patient to some risk and discomfort. Blood-based biomarkers therefore represent hope for early diagnosis and monitoring of individuals at increased risk of developing Alzheimer's disease. The aim of the diploma thesis is to determine biomarkers in the blood using ultrasensitive Simoa TM (Single molecular assay) technology and verify their relationship to the values in cerebrospinal fluid. Furthermore, the aim is to trace the relationship between blood-based biomarkers and cognitive and structural markers of Alzheimer's disease. The research group will consist of patients of the Cognitive Center of the Department of Neurology of the 2nd Faculty of Medicine, Charles University, defined according to current AT (N) diagnostic criteria (Jack et al., 2018).
