MikroRNA (miRNA) je jednou z klíčových molekul koordinujících posttranslační regulaci genové exprese. Detekce miRNA v buňkách je v současnosti klíčová pro řadu aplikací v buněčné biologii a life sciences. Ačkoliv aktuálně používané techniky jako qPCR nebo DNA čipy jsou citlivé, jsou také destruktivní a vyžadují homogenizaci buněk. Pro pochopení dynamiky miRNA v buňkách jsou potřeba vysoce citlivé optické metody, které nemají rušivý vliv na buňky. Účinnost dostupných detekčních technik může být výrazně zlepšena pomocí center dusík-vakance (NV centra) v diamantu. Elektronické a kvantové charakteristiky NV center, např. relaxační čas T1, mohou být analyzovány opticky. Diplomová práce je zaměřena na přípravu a charakterizaci nového kvantového přístupu pro detekci miRNA založeného na aktuaci spojené se sekvenčním rozpoznáním miRNA. Cyklické DNA struktury, tzv. molecular beacons, budou modifikovány a kovalentně připojeny k diamantovým nanočásticím obsahujícím NV centra. Budou studovány jejich odezvy k cílovým miRNA.

MicroRNAs (miRNA) play a huge role in cells by coordinating the post-transcriptional regulation of gene expression. Detection of miRNAs in cells has become one of key tools for cell biology and life sciences. Although the currently used techniques such as qPCR and DNA chips are sensitive, they are destructive and require homogenization of the cells. For understanding of cell dynamics of miRNA, highly sensitive non-perturbant optical methods are urgently needed. The performance of the available fluorescent detection techniques can be strongly improved using nitrogen-vacancy (NV) center embedded in diamond. Using optical means, electronic and quantum parameters such as T1 relaxation time of the paramagnetic NV center can be analyzed. The diploma thesis will be focused on preparation and characterization of a new quantum detection approach for miRNA, based on combined recognition and actuation process occurring on nanodiamond crystals. Arrays of modified DNA-based molecular beacons will be covalently linked to a diamond nanoparticle equipped with NV centers and their response to the target miRNA will be studied.
