Cílem práce je objasnění vlivu mezidoménových spojení ve vícedoménových proteinech (tzn. mezidoménové a N- a C- koncové úseky) na funkci multidoménoých proteinů. Vztah mezi složením nedoménových úseků a proteinovou aktivitou a specificitou bude studován primárně na souboru protein kináz, které patří mezi mezi nejstudovanější třídu multidoménových proteinů. V této práci bude úkolem studenta získat sekvence multidoménových proteinů obsahujících kinázovou doménu z dostupných databází (Pfam, CATH, UniProt). Seznam získaných proteinů bude studována utříděn tak, aby bylo možné vytvoři soubor rozpustných proteinů které jsou přítomné ve stejném buněčném kopartmentu. Fyzikálně-chemické charakteristiky nedoménových oblastí budou určeny pomocí teoretických nástrojů a poté budou na tomto základu proteiny klasifikovány.

This work aims to uncover how the composition of non-domain regions (i.e., inter-domain linkers, N- or C-terminal tails) in multi-domain proteins affects the function of such macromolecules. The relationship between the non-domain regions' composition and protein activity and specificity will be studied primarily on a set of protein kinases, which are among the most heavily studied classes of multi-domain proteins. In this work, the student will acquire the sequences of multi-domain proteins containing the protein kinase domain from on-line databases (e.g., Pfam, CATH, UniProt). The list of protein kinases will be explored and culled to obtain a set of soluble proteins occupying the same sub-cellular compartments. Physico-chemical features will be calculated for the non-domain regions of such set of proteins and classification based on these features will be performed.
