Genom lidských polyomavirů je tvořen kruhovou dvouřetězcovou DNA o velikosti přibližně 5 kb a lze ho rozdělit na tři funkční oblasti; časnou oblast kódující regulační T antigeny a virové miRNA, pozdní oblast kódující strukturní kapsidové proteiny a nekódující kontrolní oblast. Nekódující kontrolní oblast (noncoding control region (NCCR)) obsahuje sekvence počátku virové replikace a promotorové a regulační sekvence virové transkripce na obou řetězcích virové DNA. Regulační sekvence zahrnují řadu vazebných míst pro buněčné transkripční faktory. Cílem této bakalářské práce je detailně popsat organizaci a funkci nekódující oblasti lidských polyomavirů. Velká pozornost bude věnována zvláště regulační nekódující oblasti BK polyomaviru (BKPyV) a sekvenčním změnám, které byli identifikovány u pacientů s BKPyV asociovanou nefropatií, které vedou ke zvýšené expresi časných proteinů a replikaci viru v porovnání s archetypem BK polyomaviru.

Human polyomavirus (HPyV) genomes consist of circular ds DNA of about 5 kb in length. The genomes share the same organization and can be divided into three functional regions: the early viral gene region, encoding the regulatory T antigens and microRNAs; the late viral gene region, encoding the structural capsid proteins; and the noncoding control region. The noncoding control region (NCCR) harbor the origin of viral replication and bidirectional promoter/enhancer that control expression of early a late gene region in opposite directions. NCCR possess the numerous cells transcription factor-binding sites (TFBS). The aim of this bachelor thesis is to describe in detail the organization and function of non-coding region of human polyomaviruses. The attention will be paid to the NCCR of BK polyomavirus and to sequence rearrangement identified in patients with BK polyomavirus associated nephropathy leading to increased early protein expression and virus replication compared to nonrearranged virus.
