Přenos mutace genu Msh6 z myšího inbredního kmene C57BL/6.Msh6 (poddruh Mus musculus domesticus) do kmene PWD/Ph (poddruh Mus musculus musculus). Cílem práce je ověřit pracovní hypotézu, podle níž předpokládaný antirekombinační mechanismus mismatch repair genů Msh6 a Msh2 zabraňuje opravě „asymetrických“ PRDM9 dependentních dvouřetězcových zlomů (DSBs) DNA v meioze. Jedním z kroků k testování hypotézy je přenesení již připravené nulové Msh6 mutace do kmene PWD, což umožní přípravu PWD x B6 homozygotů pro nulovou mutaci Msh6. Karel Fusek se rovněž zúčastní dalšího kroku, který spočívá v přípravě dvojitého mutanta Msh6/Msh2, který je nezbytné generovat na pozadí s Msh6 mutací vzhledem k malé vzdálenosti obou genů, 32.2 kb na Chr 17. Pro genotypizaci mutace bude Karel Fusek amplifikovat hraniční oblasti delece pomocí PCR a metodami imunofluorescenční mikroskopie se bude podílet na zjišťování vlivu delece na četnost DSBs (DMC1) a meiotické rekombinace (MLH1) u Msh6 null heterozygotů a homozygotů na pozadí kmene B6 a u PWD x B6 hybridů.

The aim of the work is to verify the working hypothesis suggesting that the presumed antirecombination mechanism of Msh6 and Msh2 mismatch repair genes prevents repair of „asymmetric“ PRDM9-dependent DNA double-strand breaks (DSBs) in meiosis. Karel Fusek will participate also on another step to confirm the hypothesis, namely to transfer the already prepared Msh6 null mutation to the PWD strain to enable a further step, the construction of a double Msh6/Msh2 mutant. Because both genes, Msh2 and Msh6 are closely linked on Chr 17 (32.2 kb) the double mutant will be generated on background of the mouse strain with Msh6 null mutation. To genotype the mutation, Karel Fusek will amplify the sequences surrounding the deletion using PCR. He will also use methods of immunofluoresnce microscopy to study the effect of deletion on DSB rate (DMC1) and meiotic recombination (MLH1) in Msh6 null heterozygotes and homozygotes on B6 and PWD x B6 background.
