DNAs bearing reactive groups in the major or minor groove will be prepared by polymerase incorporation of modified nucleoside triphosphates.Furthermore, bioorthogonal reactions of these functional groups in DNA will be studied, especially reactions changing the size of the attached substituent for switching or triggering DNA interactions with proteins and transcription.
