Mezenchymální kmenové buňky (mesenchymal stem cells, MSCs) jsou díky svým imunosupresivním a diferenciačním schopnostem nadějným prostředkem pro transplantační i regenerativní medicínu a pro léčbu mnoha zánětlivých onemocnění. Mnoho studií popisuje použití MSCs v kombinaci s imunosupresivními látkami, ale přesný mechanismus pozorovaného synergistického působení ještě nebyl popsán. Cílem práce je analýza působení MSCs aplikovaných s imunosupresivními látkami na zánětlivou odpověď pomocí in vivo modelu transplantace alogenních buněk. Práce bude zaměřena na studium rovnováhy mezi populacíTh1, Th2, Treg, Th17 a Tc buněk, jejich aktivaci a produkci cytokinů. Výsledky získané pomocí této studie tak významně pomohou objasnit interakci mezi MSCs a imunosupresivy s různými mechanismy účinku a jejich efekt na T lymfocyty.

Mesenchymal stem cells (MSCs) possess significant immunosuppresive and multilineage differentiation capacity. Therefore they represent a promising thrapeutic tool for transplantation and regenerative medicine and for treatment of many inflammatory diseases. There are numerous studies describing the use of MSCs in combination with immunosuppressive drugs but the exact mechanism of observed synergism has not yet been described. The aim of the thesis is to analyse the effect of MSC-based treatment combined with immunosuppressive drugs on the inflammatory reaction in an in vivo model of allogeneic cell transplantation. The thesis is focused on the determination of balance among population of Th1, Th2, Treg, Th17 and Tc cells, their acivation and cytokine production. Results obtained in this study will bring substantial knowledge about the interaction of MSCs with several immunosuppressive drugs with different mechanism of action and their effects on T lymphocytes.
