Cílem této práce bude zjistit, jak aktivace μ-opioidního receptoru vybranými usměrňovacími ligandy (morfin, endomorfin 2 a DAMGO) ovlivňuje fungování a interakce receptoru s potenciálními molekulárními partnery (např. G proteiny, β-arrestin-2, GRK či RGS). Za použití fluorescenčních metod (FRAP a FLIM-FRET) bude charakterizován vliv usměrňovacích ligandů na mobilitu μ-opioidního receptoru v plazmatické membráně, na dynamiku internalizace receptoru a na jeho proteinové interakce.

The aim of this thesisi is to find out how activation of the μ-opioid receptor by selected biased ligands (morphine, DAMGO and endomorphine-2) affects the functioning and interactions of the receptor with its potential molecular partners (eg., G proteins, β-arrestin-2, GRK or RGS). The effect of biased ligands on the mobility of MOR in the plasma membrane, dynamics of the μ-opioid receptor internalization and protein interactions we will characterize using fluorescence methods (FRAP and FLIM-FRET).
