Diplomová práce se bude zabývat charakterisací dvou nedávno objevených terapeutických cílů v chřipkové RNA polymerase – doménami odpovědnými za vazbu a odštěpení čepičky hostitelské pre-mRNA. Náplní práce bude vyvinout metody využitelné k testování potenciálních inhibitorů polymerasy. Část práce bude věnována rešerši shrnující tématiku vazby a odštěpování čepičky polymerasovými podjednotkami. Během práce studentka připraví rekombinantní proteiny a využije biofyzikální metody jako např. AlphaScreen, povrchovou plasmonovou resonanci SPR či rentgenostrukturní analysu.

The master thesis will be focused on characterization of two recently identified targets in Influenza RNA polymerase – cap binding and endonuclease domains. The objective of this thesis is to develop a methods based on the AlphaSreen technology or recently published DIANA assay (Navrátil et al., 2016) that will be used for screening of potential polymerase inhibitors. The part of the thesis will be up to date review concerning cap-snatching and endonuclease activities of Influenza virus polymerase. Master student will acquire knowledge on recombinant protein expression and biophysical methods such as AlphaScreen, surface plasmon resonance (SPR) and X-ray crystallography.
