Produkce fúzního konstruktu receptoru a následná purifikace dimerní formy receptorů pomocí afinitní a HPLC chromatografie, štěpení fúzního proteinu TEV proteasou. Alternativně, příprava NKR-P1C proteinu renaturací z inkluzních tělísek exprimovaných v buňkách E. coli kultivovaných v LB médiu s induktorem IPTG anebo v autoindukčním médiu ZYP-5052. Krystalizace připravených proteinů a studium jejich struktury metodami strukturní biologie (proteinová krystalografie, hmotnostní spektrometrie, Ramanova spektroskopie, ad.). Pokus o nalezení přirozeného proteinového vazebného partnera receptoru NKR-P1C pomocí kombinace technik tandemové afinitní purifikace, imunofluorescence a síťovacích reakcí.

Preparation of dimeric soluble form of lectin receptor of mouse NK cells NKR-P1C by recombinant expression using transient transfection of HEK293 cell line. Production of receptor fusion construct and subsequent purification of dimeric receptor by affinity and HPLC chromatography, cleavage of the fusion construct with TEV protease. Alternatively, preparation of NKR-P1C protein by refolding from inclusion bodies expressed in E. coli grown in LB medium with IPTG induction or in autoinduction ZYP-5052 medium. Crystallization of the prepared proteins and their structural studies using the techniques of structural biology (e.g. protein crystallography, mass spectrometry, Raman spectroscopy, etc.). An attempt to find the natural protein binding partner of the NKR-P1C receptor by a combination of tandem affinity purification, immunofluorescence and cross-linking techniques.
