RNA interference (RNAi) hraje klíčovou roli v regulaci genové exprese a ochraně rostlinných genomů před invazivními nukleovými kyselinami. Cílem projektu je lépe porozumět mechanismu RNAi, konkrétněji indukci posttranskripčního umlčování genů (PTGS) a jeho přechodu do umlčování na transkripční úrovni (TGS). Výsledky by měly odhalit (i) závislost umlčení na původu spouštěcí dsRNA, (ii) načasování a mechanismus přechodu PTGS na TGS a (iii) role některých složek RNAi v procesu, např. RNA-dependentní RNA polymeráza RDR6. Pokusy budou prováděny s buněčnou linií tabáku BY-2, která umožňuje dlouhodobou analýzu epigenetických změn v mitoticky se dělících buňkách. Experimentální systém je založen na „indikátorových linkách“, které jsou navrženy pro snadné vizuální sledování epigenetických změn po indukovatelném spuštění PTGS. Epigenetické změny budou potvrzeny detekcí methylcytosinu po úpravě hydrogensiřičitanem.

RNA interference (RNAi) plays a key role in the regulation of gene expression and protection of plant genomes against invasive nucleic acids. The aim of the project is to better understand the mechanism of RNAi, more specifically the induction of posttranscriptional gene silencing (PTGS) and its transition into silencing at transcriptional level (TGS). The results should uncover (i) the dependence of silencing on the origin of the triggering dsRNA, (ii) the timing and mechanism of the transition of PTGS to TGS and (iii) the role of some RNAi components in the process, e.g. RNA-dependent RNA polymerase RDR6. The experiments will be performed with tobacco cell line BY-2 that allows long-term analysis of epigenetic changes in mitotically dividing cells. The experimental system is based on “indicator lines” that are designed for easy visual monitoring of the epigenetic changes after inducible triggering of PTGS. Epigenetic changes will be confirmed by the detection of methyl cytosine after bisulfite modification. The method for detection of small RNAs has to be established.
