Identifikovali jsme Arabidopsis thaliana Phosducin-like protein 2 (PhLP2) jako klíčový regulátor růstu a vývoje rostlin. Phosduciny jsou proteiny obsahující thioredoxinovou doménu, vyskytující se u všech eukaryot a o nichž je známo, že regulují signalizaci heterotrimerních G-proteinů. Úloha PhLP2 ve vývoji rostlin však zůstává neobjasněna. Dostupné údaje a naše předběžné výsledky naznačují roli PhLP2 v signalizaci G-proteinů, reakci na auxin a endomembránovém traffickingu. Tématem této práce je objasnit molekulární funkci proteinu PhLP2 tím, že budou identifikovány jeho proteinové interakce. Identita proteinových interaktorů pomůže objasnit molekulární procesy, v nichž tento protein hraje roli. V této práci student-ka využije existující transgenní linie Arabidopsis exprimující PhLP2 značené TurboID a fluorescenčními značkami k provedení metody proximity labeling, která pomáhá identifikovat proteiny v těsné blízkosti proteinu našeho zájmu. Dále bude analyzován soubor potenciálních interaktorů a vybrané proteiny budou vybrány pro další analýzu pomocí metod in silico (AlphaFold2 pulldown), in vitro a in vivo (pokročilá mikroskopie).

We identified Arabidopsis thaliana Phosducin-like protein 2 (PhLP2) as a crucial regulator of plant growth and development. Phosducins are thioredoxin domain-containing proteins found across eukaryotes and known to regulate heterotrimeric G-protein signaling. However, the role of PhLPs in plant development remains elusive. The available data and our preliminary results suggest a role of PhLP2 in G-protein signaling, auxin responses, and endomembrane trafficking. The topic of this thesis is to shed light on the molecular function of the PhLP2 protein by determining its protein interaction network. The identity of protein interactors will shed light on the pathways this protein plays a role in. In this thesis, the student will use the existing transgenic Arabidopsis lines expressing PhLP2 labelled with TurboID and fluorescent tags to perform the proximity labeling method that identifies proteins in close vicinity of the protein of interest. Further, the set of potential interactors will be analyzed, and candidate proteins will be selected for further analysis using in silico (such as AlphaFold2 pulldown), in vitro, and in vivo (advanced microscopy) methods.
