V mikroRNA dráze generuje RNáza III Dicer malé duplexy RNA, které jsou následně podrobeny selekci jednoho mikroRNA vlákna a vybrané vlákno je navázané na efektorový protein z rodiny Argonaute. Správná selekce mikroRNA vlákna je rozhodující pro kontrolu genové exprese zprostředkovanou mikroRNA. Předchozí výzkum ukázal, že termodynamická stabilita konců duplexu RNA je vnímána proteiny v komplexu přenášejícím malou RNA na Argonaut a do určité míry předpovídá selekci řetězce. Existuje však mnoho mikroRNA, které toto termodynamické pravidlo nedodržují. Pracovní hypotézou tohoto projektu je, že termodynamické vnímání konců je spojeno s 3D tvary mikroRNA duplexů a jejich orientací v komplexu přenášejícím malou RNA na Argonaut. Tento projekt bude kombinovat experimentální molekulární biologii s bioinformatickou analýzu vlastností mikroRNA prekurzorů a organizace komplexu přenášejícího malou RNA na Argonaut, aby byly nalezeny vlastnosti mikroRNA prekurzoru, které by lépe vysvětlovaly selekci správného vlákna mikroRNA než stávající model.

In the microRNA pathway, RNase III Dicer generates small RNA duplexes, which are subsequently subjected to strand selection and loading of a selected strand onto an effector protein from the Argonaute family. Proper strand selection is critical for the microRNA-mediated control of gene expression. Previous research showed that thermodynamic stability of termini of the RNA duplex is sensed by proteins in the Argonaute loading complex and predicts to some extent strand selection. However, there are many microRNAs, which do not obey this thermodynamic rule. The working hypothesis of this project is that the thermodynamic sensing is coupled with 3D shapes of microRNA duplexes and their orientation in the Argonaute loading complex. Accordingly, this project will combine experimental molecular biology with bioinformatics to analyze properties of microRNA precursors and organization of the Argonaute loading complex in order to determine features, which would better explain strand selection and loading then the existing model.
