Uropatogenní izoláty bakterie Escherichia coli produkují α-hemolyzin HlyA, protein, jenž patří mezi cytolytické RTX (Repeats in ToXin) toxiny sekretované některými gramnegativními patogenními bakteriemi. HlyA je syntetizován jako neaktivní protoxin proHlyA, k jehož akivaci dochází kovalentní acylací ε-aminoskupin dvou konzervovaných lysinových zbytků pomocí koexprimované acyltransferasy HlyC. Jako donor acylového řetězce slouží acyl-acyl carrier protein (acyl-ACP). Nicméně, molekulární mechanismus, kterým acyltransferasa HlyC interaguje s acyl-ACP a proHlyA, je prozatím jen málo prozkoumán. Hlavním cílem navrhovaného bakalářského projektu proto bude pomocí bioinformatických predikčních nástrojů a biochemických testů získat nové poznatky o interakci HlyC-acyl-ACP-proHlyA, a tím i o aktivaci protoxinu proHlyA na toxin HlyA, který je důležitým faktorem virulence podílejícím se na infekcích způsobených uropatogenní E. coli.
Předběžná náplň práce v anglickém jazyce
Uropathogenic Escherichia coli isolates secrete the α-hemolysin HlyA, which belongs to an ever-growing family of cytolytic RTX (Repeats in ToXin) toxins secreted by various Gram-negative pathogens. HlyA is synthesized as the inactive protoxin proHlyA, the central portion of which comprises two conserved lysine residues that are covalently acylated at the ε-amino groups by the co-expressed toxin-activating acyltransferase HlyC. To convert the protoxin to the active toxin, HlyC uses the acyl-acyl carrier protein (acyl-ACP) as an acyl chain donor. However, little is known about the molecular mechanism by which the acyltransferase HlyC interacts with acyl-ACP and proHlyA, and its investigation will be the central goal of the proposed bachelor project. A combination of bioinformatic prediction tools and biochemical assays will be used to provide new insights into the HlyC-acyl-ACP-proHlyA interaction and thus into the activation of the protoxin proHlyA to the HlyA toxin, which is an important virulence factor involved in uropathogenic E. coli infections.
