Náplní práce bude zavedení reportérových buněčných linií pro funkční studie interakcí NK buněčných receptorů s jejich ligandy s využitím průtokové cytometrie a sledování aktivace buněčné signalizace po vazbě bispecifických proteinů sestávajících z nanoprotilátek a ligandů NK buněčných receptorů. Konkrétně se bude jednat o reportérovou linii s extracelulární doménou lidského aktivačního receptoru NKp30 a případně také potkaního inhibičního NKR-P1B, které budou také využity pro podpůrné experimenty související s naším strukturním výzkumem těchto dvou receptorů.

The thesis aims to generate the reporter cell lines for functional studies of interactions of NK cell receptors with their ligands using flow cytometry and evaluate the activation of cellular signalization mediated by bispecific proteins composed of nanobodies and ligands of NK cell receptors. Specifically, the aim is to create a reporter cell line harboring the extracellular domain of human activation receptor NKp30 and possibly also rat inhibitory receptor NKR-P1B, which will also be utilized for supporting experiments related to our structural research of those two receptors.
