Cytotoxin RtxA je klíčovým faktorem virulence pediatricky významné gramnegativní bakterie Kingella kingae, která způsobuje septickou artritidu, osteomyelitidu, infekční endokarditidu a další onemocnění. Studium vztahu struktury a funkce RtxA a molekulárních mechanismů, jimiž toxin přispívá k patogenezi onemocnění, je proto vysoce aktuální. Náplní práce bude cílená PCR mutageneze konkrétního segmentu cytotoxinu RtxA, exprese mutantních variant v expresním systému Escherichia coli, jejich izolace, purifikace a stanovení biologických aktivit za účelem objasnění topologie RtxA v membránách hostitelských buněk.

The cytotoxin RtxA is a key virulence factor of the pediatrically important gram-negative bacterium Kingella kingae, a causative agent of septic arthritis, osteomyelitis, infectious endocarditis, and other diseases. Therefore, the study of the relationship between the structure and function of RtxA and the molecular mechanisms by which the toxin contributes to disease pathogenesis is highly relevant. The content of the work will include PCR mutagenesis of the specific segment of RtxA, expression of its mutant variants in the Escherichia coli expression system, isolation and purification of the recombinant proteins and characterization of their biological activities to elucidate the topology of RtxA in host cell membranes.
