Důležitou součástí vrozené imunity člověka jsou protivirové interferony. Jejich indukce spočívá v aktivaci signálních drah, které jsou spuštěny po rozpoznání patogenní RNA a DNA v buňce, nebo endogenní DNA nepřirozeně v buňce umístěné. Tato detekce je zprostředkována RNA a DNA senzory. Mezi nejdůležitější DNA senzory patří cGAS a IFI16, jejichž společná funkce je schopnost indukovat interferon přes centrální adaptérovou molekulu signalizace STING. Oba senzory se vyskytují v cytoplazmě i jádře buňky. cGAS plní kanonické funkce spojené s detekcí virové DNA v cytoplazmě, syntézou dinukleotidu cGAMP, který aktivuje adaptérový protein – STING. V jádře cGAS inhibuje opravy DNA a podporuje tumorogenezi. IFI16 rozpoznává genomy DNA virů v jádře, kde také působí jako restrikční faktor potlačující replikaci virů v jádře. V cytoplazmě interaguje je důležitou součástí komplexu s proteinem STING. Novější studie poukazují na nové nekanonické funkce cGAS, které se pojí k autofágii a roli IFI16 v indukci odpovědi vrozené imunity při poškození DNA.

Antiviral interferons play a crucial role in human innate immunity. The induction of interferons relies on the activation of signalization cascades which are initialized by the recognition of pathogen-derived RNA and DNA in the cell or unnaturally located endogenous DNA within the cell. This recognition depends on RNA and DNA sensors. Among the most important sensors are cGAS and IFI16 which interact with the central adapter molecule of signalling – STING. Both of these sensors are located in the cytosol and the nucleus of the cell. cGAS´s canonical functions include the detection of viral DNA in the cytosol, the synthesis of dinucleotide called cGAMP and subsequent activation of the adaptor protein STING. In the nucleus cGAS inhibits the repair of damaged DNA and therefore promotes tumorigenesis. IFI16, however, detects genomes of DNA viruses in the nucleus, where it also acts as a restriction factor therefore abrogating viral replication. In the cytosol IFI16 is an important component of a complex comprised of IFI16 and STING. Newer studies highlight the non-canonical roles of cGAS in autophagy and IFI16 in the IFI16-mediated induction of innate immune response due to DNA damage.
