Inhibitory bakteriálních a lidských adenylát cykláz mají značný potenciál při léčbě buď infekčních onemocnění nebo při léčení rakoviny, chronické bolesti, neurodegenerativních onemocnění či astmatu. Inhibitory purinových fosforibosyltransferáz mají zase potenciál při léčbě nemocí jako je malárie a trypanosomiáza. Cílem práce je návrh, syntéza a vyhodnocení biologických vlastností (SAR studie) nových typů inhibitorů těchto enzymů. Klíčovou skupinu látek tvoří strukturně modifikované nukleotidy, zejména fosfonáty acyklických nukleosidů a jejich proléčiva.

Inhibitors of bacterial or human adenylate cyclases have a great therapeutic potential in treatment of either infectious diseases or diseases as cancer, chronic pain, neurodegenerative disorders and asthma. Inhibitors of purine phosphoribosyltransferases represent agents with potential to treat diseases like malaria and trypanosomiasis. The aim of the thesis is design, synthesis and biological evaluation (SAR study) of novel types of inhibitors of these enzymes. The key group of compounds are structurally modified nucleotides, namely acyclic nucleoside phosphonates, and their prodrugs.
