Měření genové exprese je základním typem analýzy využívané při studium genomu, které poskytuje jak informace o jeho transkripční aktivitě, tak i o regulaci jednotlivých biochemických drah. Ve snaze o získání co největšího množství informací bylo vynaloženo značné úsilí ve snaze o vylepšení metod používaných při měření genové exprese. Mezi v současné době nejvyužívanější technologie se řadí metoda RNA-sekvenování (RNA-Seq) sloužící pro profilování genové exprese na celogenomové úrovni a metoda reverzní transkripce kvantitativní polymerázové řetězové reakce (RT-qPCR), která je používána pro přesné měření exprese omezeného počtu genů. Bez ohledu na zvolenou techniku je klíčovým faktorem udávající výsledek každého experimentu efektivita prvního kroku, kterým je reverzní transkripce. Cílem této bakalářské práce je vytvořit souhrn současného stavu poznání týkající se vlastností reverzních transkriptáz při kvantifikaci molekul mRNA se zaměřením na jejich aplikaci v technikách RT-qPCR and RNA-Seq. In functional genomics, transcript measurement is of fundamental importance, since it not only reveals the activity of a genome but also delivers information on the regulation of biochemical pathways. In that connection, many efforts were directed toward improving detection methods. Among the most popular technologies, the RNA-sequencing (RNA-Seq) is used for large-scale gene expression profiling, while the reverse transcription quantitative polymerase chain reaction (RT-qPCR) is applied when a precise measurement is required. Irrespective of used method, the efficiency of the initial step, the reverse transcription, is a key parameter determining the outcomes of each experiment. The aim of this bachelor thesis is to review the properties of the reverse transcriptases in mRNA quantification analysis with focus on their application in RT-qPCR and RNA-Seq technologies.
