Nanodiamanty jsou netoxické, anorganické nosiče s jedinečnými vlastnostmi a s obrovským potenciálem v biomedicínském výzkumu. Cílem diplomové práce je příprava a především aplikace těchto nanočástic k transfekci siRNA do eukaryotických buněk a sledování experimentálních podmínek vedoucích k úspěšné transfekci. Důraz bude především kladen na pozorování mechanismu vstupu nanočástice do buňky, na její osud v buňce v rámci endocytické dráhy. Dalším cílem je zhodnocení cytotoxicity nanočástic a jejich transfekční účinnosti.

Nanodiamonds are non-toxic, inorganic carriers with unique properties and with a remarkable potential in biomedical research. The goal of this work is to prepare and utilize the nanoparticles in siRNA transfection to eukaryotic cells and to establish the experimental conditions leading to a successful transfection. The nanoparticle's cellular entry mechanism and its subsequent cellular fate in the endocytic pathway will be studied. Another aim of this work will is the assessment of cytotoxic effect of the nanoparticles and their transfection efficiency.
