Diplomová práce je zaměřena na bližší charakterizaci funkce transportéru AtKUP5 v rostlině. Tento transportér je členem širší rodiny vysokoafinitních draselných transportérů AtKT/HAK/KUP, doposud však není jeho funkce v rostlině vyjasněna. Práce navazuje na experimenty ukazující odlišný fenotyp kup5 mutantů v podmínkách nedostatku draslíku (diplomová práce Marka Šustra). Cílem diplomové práce bude fenotyp inzerčních mutantů blíže charakterizovat a s pomocí transformace rostlin reportérovými geny popsat expresi AtKUP5 v rostlině a lokalizaci transportéru na buněčné úrovni i v rámci celé rostliny.

The diploma thesis is aimed on detailed characterisation of AtKUP5 plant transporter. The transporter is a member of KT/HAK/KUP family of plant high-affinity potassium transporters, but its function is unknown. The experiments are based on diploma thesis of Marek Šustr, where kup5 insertion mutants were described to exhibit different phenotype compared to wild-type plants under potassium deficiency. The present diploma thesis will therefore examine phenotype of kup5 plant (growth, potassium accumulation, response to potassium deficiency and other stress factors) and characterise KUP5 expression and KUP5 protein localization using plant transformation with reporter genes.
