Diplomová práce je zaměřena na vliv povrchové ubiquitinace prasečích spermií na jejich úspěšnost při oplození a následný časný embryonální vývoj. Těžištěm práce je izolace prasečích oocytů a jejich oplození pomocí spermií s různou mírou povrchové ubiquitinace na základě třídění metodou FACS. Oplozená embrya budou fixována a bude sledována přítomnost či absence prvojader. Dále pak budou časné zárodky analyzovány z pohledu kvality embryonálního vývoje do stadia blastocysty.

Diploma thesis is focused on the effect of sperm surface ubiquitination on their fertilization ability and subsequent early embryonic development in pig. The emphasis will be put on the isolation of porcine oocytes and their fertilization by sperm cells with the variable rate of surface ubiquitination based on the FACS sorting. Fertilized embryos will be fixed and the presence or absence of pronuclei will be observed. Subsequently, early embryos will be analyzed concerning the quality of the early embryonic development to the blastocyst stage.
