Cílem studia je poznání interakcí protein-protein a jejich specificitu ve skupině interleukinů z rodiny interleukinu 10. Koncentruje se na nejméně známé členy rodiny, tedy IL-24 a IL-26 a jejich interakce s receptory 1 a 2, zejména na mechanismus tvorby ternárních komplexů IL-receptor 1-receptor 2, které zajišťují signalizaci. Inhibice interakcí mezi interleukiny a jejich receptory by potenciálně měla velký význam pro vývoj léčebných či diagnostických prostředků. Studium se proto rovněž zaměří na vývoj nových proteinových molekul, které tyto interakce budou inhibovat. Ty budou vybrány pomocí řízené evoluce malých proteinových domén (scaffold). Termodynamické, kinetické a strukturní aspekty interakcí budou studovány pokročilými biofyzikálními technikami, sekvenční i strukturní bioinformatikou a počítačovým modelováním.
Předběžná náplň práce v anglickém jazyce
The study aims at understanding of protein-protein interactions and their specificity in the group of interleukins from the family of interleukin 10. It will concentrate on the least known members of the group, namely IL-24 and IL-26 and their interactions with receptors 1 and 2, especially mechanisms of formation of the ternary complexes IL-receptor 1-receptor 2, which enable signaling. Inhibition of interactions between interleukins and their receptors would potentially be of great medical or diagnostic interest. The study will therefore aim at developing novel protein binders inhibiting these interactions; these binders will be selected by directed evolution of small protein scaffolds. Thermodynamic, kinetic, and structural aspects of interactions will be studied by advanced biophysical techniques, sequential and structural bioinformatics, and computer modeling.
