Akutní lymfoblastická leukemie (ALL) je nejčastější maligní onemocnění u dětí. Přestože je terapie ALL velmi úspěšná, relaps onemocnění postihuje 20-25 % dětí. Jedna z možných příčin relapsu je rezistence k léčivům. Léčba leukemie spočívá v kombinované chemoterapii, jejíž důležitou součástí je L-asparagináza. Tento enzym přeměňuje extracelulární asparagin na asparát a glutamin na glutamát. Nedostatek asparaginu a glutaminu vede k poškození proteosyntézy a navození apoptózy. Detailní mechanismus působení L-asparaginázy a příčiny vzniku rezistence však nebyly dosud objasněny. Tato diplomová práce je součástí studia příčin rezistence leukemických buněk vůči L-asparagináze.

Acute lymphoblastic leukaemia (ALL) is the most common malignancy in childhood. Despite the very successful ALL therapy, relapses nowadays occur to 20-25 % of children. One of the possible relaps causes is the resistance towards therapeutics. ALL is treated with combined chemotherapy in which cytostatic agent L-asparaginase plays the essential role. L-asparaginase depletes the extracellular asparagine to aspartate and glutamine to glutamate.The asparagine and glutamine deficiency harms cellular proteosynthesis and induces apoptosis. Mechanism of the L-asparaginase cytotoxic effect and mechanism of corresponding resistance is still not fully explained. This thesis is a part of a project studying mechanisms of leukaemic cells resistance to L-aparaginase.
