Výtěžek rekombinantní exprese proteinů prováděné metodou tranzientní transfekce lidské HEK293 buněčné linie je přímo úměrný délce produkční fáze kultivace buněk. Tu je možno prodloužit cílenou zvýšenou expresí proteinů regulujících (zastavujících) buněčný cyklus a/nebo apoptosu. Cílem práce je s již připravenými regulačními plazmidy provést optimalizaci tohoto expresního systému, v první fázi s GFP a SEAP jako modelovými proteiny a následně systém ověřit pro expresi jiných cílových proteinů. Během práce bude také optimalizováno kultivační, resp. produkční médium a celkové podmínky kultivace buněčné linie během produkce.

The yield of recombinant protein expression performed by a transient transfection of human HEK293 cell line is directly proportional to the length of the production phase of the cell cultivation. It is possible to prolong the production phase by purposeful overexpression of proteins regulating (stopping) the cell cycle and/or apoptosis. Aim of the thesis is to optimize this expression system with an already prepared regulatory plasmids, firstly with GFP and SEAP as model proteins and later verify the system for the expression of other proteins of interest. Cell culture or production medium and the overall cell line cultivation conditions during the production phase will be also optimized during the work.
