NKp65 a KACL jsou dva teprve nedávno objevené lektinové receptory lidských imunocytů, které spolu dohromady tvoří receptor - ligandový pár, rozpoznávají tedy samy sebe navzájem. Oproti příbuzným párům lektinových receptorů byla pro NKp65 a KACL receptory publikována neobvykle vysoká interakční konstanta, což naznačuje, že komplex těchto dvou receptorů by mohl být dostatečně stabilní a tedy vhodný i pro strukturní výzkum pomocí proteinové krystalografie (struktura žádného příbuzného komplexu nebyla dosud vyřešena). Cílem práce je přenesení vhodného úseku genů těchto receptorů do expresních plazmidů a provedení expresního testu v HEK293 buněčné linii.
Předběžná náplň práce v anglickém jazyce
NKp65 and KACL are two only recently discovered lectin receptors of human immunocytes that form together a receptor - ligand pair, thereby recognizing each other. In contrary to other related pairs of lectin receptors, an unusually high interaction constant was published for NKp65 and KACL receptors. This suggests that complex of these two receptors might be stable enough to allow for its structural characterization by protein crystallography (structure of none of the related receptor complexes has been solved yet). The aim of the thesis is to clone a suitable receptor gene constructs into expression plasmids and their expression test in HEK293 cell line.
