Škrob je uložen v plastidech zásobních orgánů rostlin nebo v chloroplastech fotosyntetických pletiv. Tento zásobní polysacharid může být degradován hydrolyticky α-amylasami (EC 3.2.1.1.) nebo fosforolyticky fosforylasami (EC 2.4.1.1.). Extrakt připravený z listů tabáku (Nicotiana tabacum L.) štěpil barevný škrobový substrát. pH optimum hydrolytického štěpení bylo pH 8,0, pH optimum fosforolytického štěpení pH 7,4. Rychlost štěpení škrobu byla nejvyšší při teplotě 55 °C. Pro štěpení škrobu v listech tabáku má větší význam α-amylasa než foforylasa.
