Předběžná náplň práce
Oprava poškozené DNA, která je umožněna BER enzymy je zásadní pro zachování genetické informace. Tento projekt je zaměřen na odstranění 8-oxo-2′-deoxyguanosine (OG), které je důsledkem oxidativního poškození DNA. Odstranění OG báze a následné rozštěpení poškozeného DNA vlákna enzymem hOGG1 bude studováno metodami teoretického modelovaní v přímé spolupráci s experimentem. Cílem projektu je určit katalytický mechanizmus opravy DNA hOGG1 enzymem.
Předběžná náplň práce v anglickém jazyce
The repair of damaged DNA due to base-excision repair (BER) enzymes is necessary to maintain integrity of the genome in cell. The project is aimed at mechanistic study of catalytic removal of 8-oxo-2′-deoxyguanosine (OG) due to the human 8-oxoguanine DNA glycosylase 1 (hOGG1). The OG is frequently occurring and dangerous lesion due to oxidative damage of DNA. The excision of OG and subsequent scission of defective DNA strand with hOGG1 will be studied by means of theoretical calculations in close collaboration with experiment. The goal of research is to reveal the mechanistic pathway involving OG-excision that remains a mystery since the discovery of hOGG1.
