The successful completion of the first aim resulted in a publication in the RNA Biology journal. Here we unraveled the action of ncRNA BC1 in the mammalian oocyte and its effect on the rate and molecular mechanisms of regulation of the translation.

During 2022 we prepared and published a review dedicated to the role of polo-like kinase 1 (PLK1) in mitosis and meiosis with the focus on mammalian oocyte.

Completion of the 2nd aim resulted in the presented publication. Here we found that novel lncRNA is expressed in several splicing forms one of which is oocyte-specific. Oocyte-specific form of Rose lncRNA is important for oocyte cytokinesis and during embryo cleavage after ZGA in a 2-cell embryo. Therefore, we have shown that previously unannotated lncRNAs expressed in the mammalian oocyte might play important roles in its and future embryo development.

In this work, we examined the expression and localization patterns of various non-coding and coding RNAs and develop a novel approach for visualization of a single mRNA together with its translational event. We analyzed the expression and localization of selected ncRNA target IAPLtr1a. Using RNA FISH we observed that IAPLtr1a ncRNA is present in the nucleus of a fully grown GV oocyte and after NEBD become dispersed in the cytoplasm on the MII stage. After fertilization IAPLtr1a becomes almost absent in the zygote.
