The original goal of the project - the ex vivo expansion of HSPCs via b-catenin-TCF/LEF inhibition - was not achieved. In search for the general function of b-catenin-TCF/LEF signaling in the hematopoietic system, we discovered that this signaling pathway directly upregulates G-CSF-R through which it promotes neutrophilic differentiation of HSPCs. Consistently, loss of b-catenin-TCF/LEF signaling resulted in reduced neutrophil counts and enhanced susceptibility of experimental mice to the infection. These results were published in the attached publication.
