Light-matter interaction at the micro- and nano-scale level. Fourier optics and diffraction limit. Fundamentals of conventional light microscopy and imaging. Fluorescence and confocal microscopy. Super-resolution microscopy and structured illumination microscopy. Non-linear and two-photon microscopy. Evanescent wave microscopy and near field optical microscopy. Light-sheet microscopy and 3D imaging techniques.

This course aims at modern methods in optical microscopy. Building from fundamental principles we reach to a survey of state-of-the-art imaging techniques addressing their intrinsic and technical limitations and major fields of applications.

The oral exam is based on the lectures and provided supplementary materials. Prior to the exam student will receive five recent publications related to the scope of the lecture. The second part of the oral exam will focus on one of the publications.

1) Průřez mikroskopickými technikami od konvenční mikroskopie k nano-optice.

2) Základy optického zobrazování. Absorpce, rozptyl a fluorescence. Citlivost a rozlišení v zobrazování.

3) Fourierovská optika a difrakční limit. Fluorescenční mikroskopie, konfokální mikroskopie, mikroskopie strukturovaným osvětlením.

4) Mikroskopie v blízkém poli: rastrovací zobrazení a zobrazení v širokém zorném poli.

5) Super-resolution fluorescenční mikroskopie v dalekém poli.

6) Optická mikroskopie ve 3D. 3D super-resolution metody. Současné aplikace v biologii.

7) Moderní výzkumné směry v optické mikroskopii. Mikroskopie napříč vědními obory.
