The lecture is designated for pre gradual and post gradual students specialized on analytical chemistry. In the introductory part, the students are get acquainted with fundamental construction parts of mass spectrometer and common instrumentation used in analytical practice. The following part deals with the fundamentals of the interpretation of mass-spectra of organic compounds. Individual fragmentation mechanisms are explained and discussed in connection with real electron impact mass spectra.

Introduction. Experimental setup of mass spectrometer. The survey of ionization techniques. Mass analyzers. Magnetic sector. Electric sector. Quadrupole mass analyzer. Ion trap. Time of flight mass analyzer. Detectors. Multiplier detectors. Scintillation photomultiplier detectors.

Mass spectrum. Molecular ion. Analysis of isotopic composition. Stable isotopes. Ring + double bonds rule.

Theory of fragmentation. Charge and unpaired electron localization in molecular ion. Fragmentation of molecular ions, fragmentation pathways. Factors influencing ion intensity. Fragmentation mechanisms of organic molecules. Sigma bond cleavage. Alpha cleavage initiated by radical site. Cleavage initiated charge site. Cleavage of cyclic structures. Rearrangement reactions. Rearrangements initiated by radical site. Rearrangements initiated by charge site.

Mass spectra of simple organic compounds and their interpretation.
