Teaching of Pharmacology in this initial part is focused on the introduction to basic principles (General Pharmacology) with consequent education of special pharmacotherapeutic chapters united to topics according particular organ systems.

Credit

The attendance on practical classes is compulsory (only one absence is acceptable). The average from all classifications in the term (semester) must be below or equal to 3.00 In the case of failure to meet the conditions for the credit: The case of two absences - the student will be examined in depth from the topics of both practical classes on which the student was absent. The case of three or more absences - the student will be examined from the topics covering the whole term (semester). The case of the average grading > 3.00 - the student will be examined from the topics covering the whole term (semester).

  1. Basic principles of pharmacology. Pharmacodynamics. The seminar on: Nature of drug targets, receptors and their types. Well-established second messengers. Dose-response curve. Potency and efficacy. Agonists and antagonists. Up and down regulations. Variation in drug responsiveness. Practical demonstrations: PC simulation - Isolated ileum stimulation. Drugs available: acetylcholine, atropine, papaverine, physostigmine, carbachol, methacholine. Topics: Agonists and antagonists. Dose- respose curve construction and its modification. The demonstration will be performed by the lecturer.
  2. Pharmacokinetics of the drugs. The seminar on: Main mechanisms determining pharmacokinetics of a drug (physico-chemical properties, permeation through biomembranes, binding). Absorption, distribution, metabolism, excretion (ADME). Pharmacokinetic parameters, pharmacogenetics, bioavailability, bioequivalence. Practical demonstrations: PC simulation using the PkSims software. Practical demonstration of the influence of the route of administration and dosing regimen on the pharmacokinetics. Calculation of bioavailability. Pharmacokinetics after continuous and continuous-intermittent dosing. Linear and nonlinear pharmacokinetics. The demonstration will be performed by the lecturer.
  3. Adverse and toxic effects of drugs. Drug interactions. Drug development. The seminar on: the above given topics, drug information and its sources. Practical demonstrations: AISLP (Electronic version of the Czech Drug Formulary). The demonstration will be performed by the lecturer.
  4. Drugs affecting cholinergic transmission: cholinergic agonists & antagonists. The seminar on: the above given topic. Practical demonstrations: PC simulation - Cardio heart rate and blood pressure (in vivo). Drugs available: acetylcholine, atropine, neostigmine, hexamethonium. The demonstration will be performed by the lecturer.
  5. Drugs affecting adrenergic transmission: adrenergic agonists & antagonists. The seminar on: the above given topic. Practical demonstrations: PC simulation - Cardio heart rate and blood pressure (in vivo). Drugs available: adrenaline, noradrenaline, isoprenaline, prazosine, propranolol, atenolol, cocaine, tyramine. The demonstration will be performed by the lecturer.
  6. Control of pain - seminar. Basic and clinical pharmacology of analgesics, both opioid and non-opioid (including NSAIDs!). Case report: Postoperative pain in children.
  7. Neuroleptic, antidepressant agents, anxiolytics - microlesson and seminar. Video recording: Extrapyramidal syndrome due to neuroleptics.
  8. Hypno-sedatives, antiparkinsonics and antiepileptics - microlesson and seminar. Practical demonstrations: Therapeutic drug monitoring (TDM) in antiepileptic therapy.
  9. Skeletal muscle relaxants. General and local anesthetics. The seminar on: the above given topic. The Credit.

Notes

  1. Students are supposed to be theoretically prepared on each practical lesson/seminar and the understanding to the topic will be regularly examined (MCQ tests/written tests/oral examination).
