Annotation -
The aim of this course is to introduce students to experimental methods for the isolation and in vitro culture of mouse oocytes and early embryos. Students will gain practical experience with live-cell microscopy techniques, enabling them to directly observe proper oocyte and embryo development. The course also includes hands-on work with real data obtained from imaging of live mouse oocytes by using confocal and light-sheet microscopy, focusing on both qualitative and quantitative analysis of meiotic maturation. Additionally, the course presents advanced microscopy techniques, including confocal and light-sheet microscopy, and discusses the advantages and limitations of different approaches for imaging live oocytes and early embryos.

The primary language of the course is Czech, but if at least one student requests, the course will be in English.

Course assessment methods and requirements for successful completion, grading scheme -

Active participation in the course and the continuous completion of assigned questions and tasks to assess practical skills acquired during course.

Last update: Drutovič David, RNDr., Ph.D. (02.02.2026)
Syllabus -

The practical course will take place over four days (Tuesday to Friday) in May or June; the exact dates will be announced. A practical demonstration of confocal and light-sheet microscopy will be held on the final day of the course at the Institute of Animal Physiology and Genetics of the Czech Academy of Sciences in Liběchov.

Task 1: Analysis of early mouse embryo development up to the blastocyst stage

hormonal stimulation of mice to obtain early embryos, isolation and culture of two-cell stage mouse embryos, and continuous monitoring of embryo development

Task 2: Experiment to study the role of a protein in meiotic maturation of mouse oocytes

isolation of mouse oocytes in prophase I, in vitro culture of oocytes to metaphase II, and use of pharmacological inhibitors

Task 3: Processing microscopy data and basic image analysis

processing of raw data, preparation of publication-quality images and movies

Task 4: Analysis of experimental data from fluorescence live-cell microscopy of mouse oocytes

analysis of time-lapse imaging of spindle assembly, securin degradation, APC activation, and chromosome segregation

Task 5: Practical demonstration of confocal vs. light-sheet microscopy

visualization of chromosomes and spindle in oocytes, use of fluorescent proteins and the fluorogenic probe SiR-tubulin, live-cell imaging

The course is taught with the support of the project reg. number CZ.02.2.69/0.0/0.0/16_015/0002362

Learning outcomes -

Upon successful completion of the course, students will be able to:

  1. Explain the principles of hormonal stimulation of mice and justify its use for obtaining oocytes at the prophase I stage.
    (Understanding)
  2. Apply appropriate laboratory procedures for the isolation of mouse oocytes at the prophase I stage and follow the principles of working with biological material obtained from laboratory animals.
    (Application)
  3. Perform in vitro oocyte culture and monitor their progression from prophase I to metaphase II.
    (Application)
  4. Describe the cellular and morphological changes accompanying meiotic maturation of oocytes during culture.
    (Understanding)
  5. Use fluorescent probes and fluorescent proteins to analyze meiotic maturation of oocytes and visualize chromosomes and the meiotic spindle in living cells.
