A key application of molecular cytogenetics is the visualization of specific sequences on chromosome
preparations or interphase nuclei using Fluorescence In Situ Hybridization (FISH). The FISH method has a wide range of applications in research and diagnostic applications. Students will have the opportunity to perform FISH on chromosome slides with several probe variants (whole genome probes, tandem repeat probes) as well as with different types of probe labelling (direct labelling with fluorescent dyes, biotin labelling and subsequent detection with fluorochrome-associated antibodies). Participants will go through the procedure from chromosome preparation, performing FISH and subsequent microscopic evaluation, image postprocessing and presentation. I. Introductory seminar-about one week before the practical part of the course. II. Preparation of slides, evaluation of meiotic division on Giemsa stained slides. III. FISH I, pre-fitting slides, FISH deployment, probe preparation and labelling. IV. Excursion to AVČR Liběchov, laser microdissection of chromosomes, demonstration of automated systems for cytogenetic evaluation (Metafare, Ikaros). V. FISH II slide washing, signal detection and amplification of biotin-labeled probes, slide finalization. VI. Individual/pairwise evaluation of slides under fluorescence microscope. VII.
