The subject has a theoretical and practical part. The theoretical part focuses on a definition of the basic principles of the cuticular analysis, history, and shows a basic overview of the leaf anatomical structure, techniques of preparation and methods of leaf maceration based on use of the evergreen and deciduous leaves. Basis of the terminology and topography in epidermal characteristics (i.e., idiocuticular and epicuticular features) including a morphological classification of stomata, trichomes and hypodermic objects are presented. The practical part is focused on a demonstration of the main epidermal features mainly by gymnosperms and angiosperms including their detailed scientific descriptions, practical preparation and determination of the modern and fossil samples.

Předmět se dělí na teoretickou a laboratorní, praktickou část. V teoretické části předmětu jsou definovány principy kutikulární analýzy, její historie, jednotlivé typy kutikulární analýzy, rekapitulována anatomická stavba listu, kutikuly sensu stricto a kutikula sensu lato. Dále jsou podrobně probrány jednotlivé způsoby preparačních technik a metod macerace kutikul. Teoreticky je charakterizována terminologie a topografie s ohledem na idiokutikulární a epikutikulární znaky včetně klasifikace stomat a trichomů a hypodermálních útvarů. V praktické části jsou nejprve demonstrovány jednotlivé typy kutikul recentních a fosilních zástupců vybraných skupin vyšších rostlin včetně podrobného popis kutikulárních struktur. Dále jsou vedeny preparace a macerace fosilních a recentních vzorků s následným vyhotovením trvalých preparátů.

Obsah kurzu:

Přednášky:

1. Definice a principy kutikulární analýzy a její historie, typy kutikulární analýzy (hromadná analýza, analýza fragmentů, analýza kutikul in situ), anatomická stavba listu, kutikula s.s., kutikula s.l., preparační techniky, peelingová metoda.

2. Terminologie: Idiokutikulární znaky - tloušťka, skulptura, papily; Epikutikulární znaky - normální epidermální buňky (obrys, antikliny), typy trichomů a jejich báze, typy průduchů a jejich klasifikace (polocytní, tetracytní, paracytní, anisocytní, diacytní, pericytní, desmocytní, copericytic, amphipericytic), hydatody, hypodermis, idoblasty a vnitřní sekreční orgány(skleroidy, pryskyřičné kanálky, sekreční nádržky); Topografie: orientace, rozmístění a hustota jednotlivých epikutikulárních znaků (hustota průduchů, stomatární index).

Cvičení:

3.-9. Praktická demonstrace a popis kutikulárních struktur u vybraných skupin vyšších rostlin.

10. Preparace a zhotovení trvalých preparátů.
