Microscopic techniques is an exercise aimed to introduce students to various types of microscopy (light microscopy, fluorescence and electron microscopy) and to practical use of these techniques on model examples (counting of micoscopic objects, digital image analysis).
The course will take place in 4 independent classes, 4 hours each. Students should enlist for the classes acording to their time possibilities. Během zápočtového týdne budeme udělovat zápočty v těchto termínech: středa 11.1. 14:00-16:00 hod. pátek 13.1. 10:45-12:00 hod.
vždy v Parazitologické posluchárně (V7, 1. suterén). K zápočtu je nutné předložit protokoly z 1., 3. a 4. bloku cvičení. Další hromadný termín/ny ve zkouškovém oznámím později.

1. Light microscopy (Hampl et al.) Introduction to the basic theory of light microscopy and handling the light microscope (oil immersion, phase contrast, Nomarski differential interference contrast). Students will prepare native and stained microscopic slides (Giemsa-Romanowski staining, opal blue and native carmin staining). The techniques will be demonstrated on protozoa with various taxonomic affiliation (trichomonads, trypanosomes, ciliates and amoebae).
2. Special light and fluorescence microsopy (Hampl et al.) Students will be introduced into the basics of the theory of contrasts in the light microscopy (phase contrast, DIC, Hofman contrast), fluorescence and confocal microscopy. They will practically learn the work with phase contrast and fluorescence microscope including the preparation of specimens. During the course we will visit the specialised laboratory of confocal microskopy.
3. Electron microsopy (Sacherová) Students will be introduced into the theory of electron microscopy and preparation of specimens. They will have chance to work practically with scanning and transmission electron microscope and prepare simple specimens.
4. Introduction to the image analysis (Jana Albrechtová, Aleš Soukup et al.) In this section, students learn the basics of digitization and image processing, phases and basic operations of image analysis. Practical exercises are conducted in the program ImageJ (National Institutes of Health, Bethesda, Maryland, USA, URL: http://imagej.nih.gov). More on the website of Block 4.
