Annotation -
The course provides principles of decay of radioactive compounds, the knowledge about different types radiation and some specific radioisotopes. The students are prepared for work with isotopes in theory as well as in practice. They are informed about the low, rules, risks, and responsibilities they have to respect when handling isotopes. They are taught to handle the isotopes properly doing several model experiments.

100% účast na přednášce a praktických cvičeních, zpracované protokoly z experimentů, znalost teoretického podkladu experimentů. Znalost bezpečnostních pravidel pro práci s radioizotopy.

Syllabus -

1) Theoretical part: Types of radiation, principles of scintillation, spectrometry and other techniques. The radioisotopes in biological experiments: characteristics, rules, risk, low, and responsibilities. Measurement of radioactivity and monitoring the radiation exposure by using personal dosimetry. Biological background of radiation effects and protection.

Practical part:

1) Synthesis of bacterial DNA a RNA in vivo: Measurement of the involvement of inhibitors of proteosynthesis by using double-label methodology (14C, 3H).

2) Measurement of the (Na+, K+)-ATPase in red blood cells: Monitoring of the flow of rubidium ions 86Rb+ into the cells in the presence/absence of specific inhibitor, ouabain.

3) Synthesis of DNA in vitro: Nick-translation (32P).

4) Construction of calibration curve for 14C.
