The course includes practical examples and theoretical background of a selection of modern methods of plant cell and molecular biology, in particular the techniques employed to study gene expression in plant material, transient expression of introduced genes in plant cells and protoplasts, and construction and analysis of transgenic plants.

Ověření výstupů učení: Learning outcomes – practice Course: Practicum in plant cell and molecular biology - MB130C34 After completing the practical exercises, the student: • explains and applies the basic principles of plant cell biology – the function of the nucleus in gene expression, the importance of the cell wall for maintaining the function and shape of cells, the function of proteins of the endomembrane system and cytoskeleton. • learns and performs methods - transformation and gene expression, preparation and in vitro cultivation of protoplasts, cellular localization of proteins, observation of the cytoskeleton, confocal laser microscopy and quantitative evaluation of cell structures, preparation and electrophoresis of RNA and proteins, quantitative RT-PCR – all that during laboratory experiments. • obtains, processes and analyzes experimental data • formulates hypotheses, interprets results and critically evaluates them with regard to experimental limitations and theoretical knowledge acquired in lectures. • integrates experimental results into a structured laboratory protocol and formulates substantive conclusions. • cooperates with colleagues in laboratory work and adheres to the principles of good laboratory practice. Verification of learning outcomes: Active participation in practical exercises and continuous submission of laboratory protocols documenting the achievement of learning outcomes and their connection to the theoretical knowledge of the course.
